The Becker group has developed a robust protocol to generate cerebellar neurons and organoids from human induced pluripotent stem cells (iPSCs). This allows the study of early developmental cerebellar physiology and pathophysiology. Combining this with CRISPR/Cas9 genome editing, we hope to recapitulate the formation of medulloblastoma in the developing cerebellum. Medulloblastoma (MB) is the most common malignant pediatric brain tumour and has several well established molecular sub-groups. Creating a model of medulloblastoma formation in cerebellar organoids would allow the study of early tumorigenisis and could serve as an advanced model for therapy testing.
