Adaptive optics for 3D structured illumination fluorescence microscopy

© 2018 The Author(s). Structured illumination microscopy (SIM) is an established technique for observing live cells and tissues at resolution that is almost twice that of the conventional microscopy. However, super-resolution images are more susceptible to negative effects from optical aberrations. Furthermore, distorted SIM fringes can lead to imaging artefacts and failed reconstructions. Here we present an adaptive optics enabled three-beam interference 3D SIM fluorescence imaging setup. Adaptive control of wavefront for aberration control and remote focusing is enabled by novel hybrid wavefront sensing approach that combines interferometric sensor and image quality driven sensing.
